It has been shown that increasing low fiber high fermentable carbohydrate diets increase the ruminal lipopolysaccharide (LPS) derived from gram negative bacteria cell wall and disturbs the reproduction performance. The aim of this study was to determine the effect of LPS on oocyte maturation and subsequent development in vitro. Accordingly, ovine cumulus oocyte complexes (COCs) were matured in medium supplemented with LPS or in the same medium without LPS (control). Nuclear maturation, cleavage and blastocyst rate, mitochondrial membrane potential, intracellular reactive oxygen species (ROS) content and Changes in transcript abundance were evaluated. Method of 3 tag digital expression gene profiling revealed that 7887 genes were expressed and only eight genes showed changes in transcript abundance. The down regulated genes included (Tripartite motif containing 25 (TRIM25), Tripartite motif containing 26 (TRIM26), Zona Pellucida glycoprotein 3(ZP3), family with sequence similarity 50-member A (FAM50), Glyoxalate and hydroxy pyruvate reductase (GRHPR), NADH ubiquinase oxireductase subunit A8 (NDUFA) and data showed the expression of cornichon family AMPA receptor auxiliary protein 4 (CNIH4) tend to be down regulated, while our data showed up regulation for Centrin 3 (CETN3).In conclusion, these findings show that LPS has undesirable effects on the maturation competence of ovine oocyte and subsequent embryo development. In addition, transcriptome profiling results can shed light on molecular mechanisms of LPS-induced infertility in ruminants.
