We used DNA meta-barcoding methods to analyse the biodiversity in 56 soil samples collected along an elevation gradient on a temperate forested island (Hauturu, New Zealand). We amplified and sequenced six barcode loci (prokaryote 16S, eukaryote 18S, fungal 26S and ITS, and two regions of the animal COI gene) with the aim of comparing patterns of biodiversity and distribution among the different taxonomic groups detected.
