We aim to elucidate the functional effects of PGE2 and non-coding, inflammatory bowel disease (IBD)-associated genetic variants on T cells exposed to activation stimuli and other inflammatory mediators that are known to be overexpressed in IBD tissues and to influence the development, function, and plasticity of T cells, particularly Th17 cells. Using DOGMA-seq, a single-cell multi-omics approach, we profiled 171,829 T cells isolated from the peripheral blood of 25 healthy adult study subjects with ages 18-35 years at the time of recruitment. All participants were enrolled with informed consent under a research protocol approved by the University of Pittsburgh Institutional Review Board.
