<br>Obesity represents a significant health challenge worldwide, with an increasing trend and high prevalence in Mexico. This metabolic disease, characterized by an increase in body mass index (BMI), leads to abnormal fat accumulation that contributes to several pathologies. The risk of obesity depends on environmental factors and diverse biological factors, such as single nucleotide polymorphisms (SNPs). Population genetic studies have demonstrated that genetic admixture plays a key role in the prevalence and expression of six SNPs associated with obesity. This pilot study investigated the prevalence and genetic frequencies of six SNPs in genes, <i>FTO</i>, <i>MC4R</i>, <i>DRD2</i>, <i>FABP2</i>, <i>ADRB2, </i>and <i>SH2B1</i>, that were previously associated with obesity and metabolic problems, in a sample population (n=129) from Mexico City. The genotyping results revealed significant variations in allelic frequencies across the analyzed polymorphisms, with SH2B1 rs4788102 showing the highest alternative allele frequency (0.41), followed by ANKK1/DRD2 rs1800497 (0.32) and FTO rs9939609 (0.31), whereas MC4R–RNU4-17P rs17782313 presented the lowest frequency (0.12). <i>U</i>nderstanding the prevalence of these genetic markers related to obesity may support the development of personalized prevention strategies and contribute to a deeper insight into the molecular basis of metabolic diseases in Mexico.
