The goal of this study is to determine the oviduct responds to 17beta-estradiol (E2) in an in vivo environment using mouse models Overall design: Adult female mice (C57B6/J, 8-12 weeks old) were purchased from the Jackson (JAX) laboratory. Animals were ovariectomized and left for 14 days to recover and eliminate endogenous steroid hormones . Sesame oil (100 uL) was used as a negative (Vehicle or Veh) control group. 17beta-estradiol (E2) was diluted in 100 uL of 10% ethanol and sesame oil. Ovariectomized females were subcutaneously injected with Veh or E2. Twenty-four hrs after injection, the oviduct was collected for scRNA-seq analysis. Vaginal washes were collected from adult female mice (between 8-12 weeks old) to determine the stage of estrous cycle. At 11:00 h, oviducts from females in estrus were collected for subsequent scRNA-seq analysis.
