Our quantitative RT-PCR analysis suggested a significant inverse relationship between PPARa activation and let-7 expression in mouse liver. Phenotypic analysis of hepatocyte-specific let-7b/c2 knockout (let7b/c2 LKO) mice showed significnat less weight gain after high-fat diet (HFD) feeding. To discover potential clues for the physiological role of let-7 miRNA in liver, RNA-seq analysis was performed in let-7b/c2 LKO mouse mice fed a HFD. Our data revealed that Let-7 miRNA disruption in hepatocytes resulted in suppression of PPARa signaling.
