RNA-seq analysis of Pten's function in regulating embryonic stem cell transition from naïve to primed pluripotent state

The goals of this study are to explore the function of Pten in regulating the pluripotency transition of mouse embryonic stem cells through Pten knockout and mutation. Overall design: we generated Pten-/- and Pten-A3 mutant mouse ESCs with the CRISPR-Cas9 system and explored the effects on the pluripotency transition of mouse embryonic stem cells. We used a series of biological technologies such as Western blot, quantitative real time PCR, and cell culture to verity the conclusions.
