we first develop uCoTarget, utilizing a split-pool barcoding strategy for realizing ultrahigh-throughput single-cell joint profiling of multiple epigenetic proteins. We then develop uCoTargetX, an expansion of uCoTarget to simultaneously measure transcriptome and multiple epigenome targets. Overall design: K562 and Kasumi cells were colleced and submited to uCoTarget for co-profiling H3K4me3 and H3K27me3. Different T5 barcodes were used to identify K562 and Kasumi, which were then splitted and pooled for 4 rounds to realize single-cell resolution. H1 and K562 Cells were colleced and used for uCoTargetX experiments.
