Lipofuscin is known as intracellular waste substance produced by damaged mitochondrial incompleted decomposition. And it is a biomarker of aging. As aging, accumulation of the lipofuscin in retinal pigment epithelium induces blindness called Age-related macular degeneration. Soraprazan is developed to reduce the lipofuscin production and cure the age-related macular degeneration. Soraprazan reducing the age biomarker lipofuscin can be suggested to have effect of longevity extension. C.elegans were treated on NGM agar plate containing 0, 50, 100, 200 M soraprazan and showed the lipofuscin reduction and extension of longevity. To identify the mechanism, 0 and 200 M treated worms RNA expression was analyzed through microarray.
脂褐素(Lipofuscin)是由受损线粒体未完全分解产生的细胞内代谢废物,同时也是衰老的生物标志物。随着机体衰老,脂褐素在视网膜色素上皮(retinal pigment epithelium)中的积累会引发名为年龄相关性黄斑变性(Age-related macular degeneration)的致盲病症。索拉赞(Soraprazan)的研发旨在减少脂褐素生成并治疗年龄相关性黄斑变性。鉴于索拉赞能够降低衰老生物标志物脂褐素的水平,其被认为具备延长寿命的潜在功效。研究人员将秀丽隐杆线虫(C.elegans)接种于含0、50、100、200 M索拉赞的NGM琼脂平板(NGM agar plate)上培养,结果显示受试线虫体内脂褐素水平降低且寿命得以延长。为探明其作用机制,研究人员通过基因芯片(microarray)技术对经0 M与200 M索拉赞处理的线虫进行了RNA表达谱分析。
