Effect of TDNC1 ectopic expression on global gene expression pattern. Homo sapiens

Type 2 Diabetes (T2D) is a complex metabolic disorder due to a progressive insulin secretory defect on the background of insulin resistance. We found a muscle specific lncRNA we named TDNC1(T2D down-regulated non-coding RNA 1) whose expression is reduced in T2D patients as well as young individuals with family history of T2D. We used Microarray to assess for global gene expression pattern following ectopic expression of this lncRNA. Overall design: Inducible flp in HEK293cells stably expressed TDNC1 along with random control RNA were created using Flp-In T-Rex Core kit according to manufacture protocol (Invitrogen). Stable clonies were pooled and RNA was extracted using Trizol and Qiagen Rneasy columns. Total RNA was hybridyzed to Affymetrix Human Gene ST 2.0 array
