Transcriptomic analysis of androgen responsive LNCaP cells depleted (RPS6KB1 KO), overexpressing for RPS6KB1 (RPS6KB1 OE) and treated with berberine (24h treatment with 5 microg/ml prior to radiation) in combination with radiation. Results provide insight into the role of RPS6KB1 in response to radiation. Logarithimically growing parental, RPS6KB1 KO and RPS6KB1 OE LNCaP cells cells (2 10 cm dishes per sample in triplicate) at 70-80% confulency were harvested for preparation of total RNA for use in transcriptomic analysis. LNCaP cells pre-treated with vehicle control (methanol) or berberine (5 microg/ml) for 24h were exposed to ionizing radiation (2Gy for 6h). In addition cells exposed to radiation alone (2Gy) were also included. Cells were harvested for total RNA preparation 6h post-radiation. Additionally, to identify transcripts shared between berberine treatment and RPS6KB1 depletion, transcriptomic data from RPS6KB1 depleted cells and was integrated with LNCaP cells treated with berberine.
