Optimisation of DNA extraction from nasal lining fluid

The aim of this study was to optimise unbiased DNA extraction from nasal lining fluid for assessing the nasal microbiome. Samples were obtained from healthy volunteers using nasosorption devices or nasal swabs, or were microbial standards of known composition from a commercial supplier (Zymo Research). Microbial profiling was performed by amplifying the V3/4 region of the 16s rRNA gene using an Illumina MiSeq, amplifying the entire 16s rRNA gene using the PacBio Sequel II, or a fragment of 16s/ITS/23s using the Oxford Nanopore MinION.
