Single cell RNA-seq of Nutlin3a-Induced Senescent mouse embryonic fibroblasts with or without expressing OSKM

Purpose: Use scRNA-seq to determine the trajectories of transcriptomes during partial reprogramming of nutlin-3a senescent MEFs. Methods: MEFs were isolated from p16-CreERT2/rtTA3-mKate2/TRE-OSKM-mCherry mice. After the senescence induction through RO3306 and nutlin-3a, treatments with 4-OHT for 9 days and doxycycline for 12 days were performed to induce OSKM expression. The doxycycline treated MEFs and untreated MEFs were collected by trypsinization and directly used for scRNA library preparation through droplet system. Examining differentially expressed genes and identifing transposable elements within quiescent and nutlin3a-induced senescent pulmonary fibroblasts
