Goal: The aim of the project was to investigate whether S.aureus proteins SPIN and 6PGD via the formation of the hybrid auto-antigen SPIN:MPO or MPO mimitope 6PGD could explain the differential aetiologies of the disease GPA (Granulomatosis with polyangitis) caused by MPO-ANCA (Anti Neutrophil Cytoplasmic Autoantibodies).Methods: Isolation kinetic verification of S.Aureus 6PGD and SPIN by means of Activity measurements, see Readme for further specifications. Determination of antibody reactivity/titers using ELISA in sera from patients with MPO ANCA and healthy individuals against the hybrid auto-antigen MPO:ANCA or 6PGD, see Readme for further specifications. Data: Antibody titers ELISA and (Kinetics) colorigenic activity assays.<br>

研究目标:本项目旨在探究,金黄色葡萄球菌(Staphylococcus aureus)蛋白SPIN与6PGD通过形成杂交自身抗原SPIN:MPO,或是MPO模拟表位6PGD,能否解释由MPO-ANCA(抗中性粒细胞胞浆自身抗体,Anti Neutrophil Cytoplasmic Autoantibodies)引发的GPA(肉芽肿性多血管炎,Granulomatosis with polyangitis)的差异化病因学机制。 研究方法:通过活性测定法完成金黄色葡萄球菌6PGD与SPIN的分离动力学验证,详细参数详见自述文件(Readme);采用酶联免疫吸附试验(ELISA,Enzyme-Linked Immunosorbent Assay)检测MPO-ANCA阳性患者与健康个体血清中,针对杂交自身抗原MPO:ANCA或6PGD的抗体反应性与抗体滴度,详细参数详见自述文件(Readme)。 数据集内容:包含抗体滴度ELISA检测结果及(动力学)显色活性测定结果。
