An experimental dataset was generated from the directed evolution of Thermococcus kodakarensis DNA polymerase (KOD DNAP). The sites (R406, S407, L408, P410, L457, R460, K487, I488, N491, D540) were targeted for saturation mutagenesis. Libraries were selected through Compartmentalised Self-Replication (CSR) for the engineering of variants capable of 2' fluoropurine incorporation. Selections were also carried out with dNTPs to additionally test the mutational impact on DNA synthesis.
