RNA-seq analysis of the impact of intermediate passaging on cell fate determination of mesendoderm progenitors

To investigate the impact of intermediate passaging on cell fate determination, cells were passaged during mesendoderm differentiation, and samples collected at different time points were subjected to bulk RNA-seq. We report here that intermediate passaging alters cell fate and directs mesendodermal progenitors toward cardiac cell fate. Overall design: Bulk RNA-seq on samples collected on Day 0,1,2,3,5,7,10 of differentiation, comparing non-passaged cells differentiated in the presence and absence of WNT inhibitor versus cells that differentiated following intermediate passaging.
