we first develop uCoTarget, utilizing a split-pool barcoding strategy for realizing ultrahigh-throughput single-cell joint profiling of multiple epigenetic proteins. We then develop uCoTargetX, an expansion of uCoTarget to simultaneously measure transcriptome and multiple epigenome targets. K562 and Kasumi cells were colleced and submited to uCoTarget for co-profiling H3K4me3 and H3K27me3. Different T5 barcodes were used to identify K562 and Kasumi, which were then splitted and pooled for 4 rounds to realize single-cell resolution. H1 and K562 Cells were colleced and used for uCoTargetX experiments.
