We studied the evolution of gene expression in Tribolium castaneum after beetles' exposure to a new environment that differed from ancestral control conditions in temperature and humidity. We used the Tribolium castaneum Cro1 strain collected from a wild population in 2010 and adapted to lab standard conditions (33°C, 70% relative humidity) for more than 20 generations. The samples originate from a paternal half-sib cross with a split brood design whereby full-sib families were split between two conditions, the control (CT: 33°C, 70% r.H.) and the Hot-Dry (HD: 37°C, 30% r.H.) conditions. Sampled individuals were reproducing females at the age of eight weeks after. Individuals were the F1 brood produced by the paternal half-sib cross. This sample consist of 8 females from 48 full-sib families with 4 females raised in CT and 4 females raised in HD conditions per full-sib family. We performed full-body RNA extraction with RNA Mini Prep kit (Zimo Research, California, USA). mRNA was used for library preparation (using LEXOGEN kit) and single end sequencing was performed on Illumina NextSeq (75 cycles).
