We developed a genetic mouse model of chronic necroptosis activation in hepatocytes (in the presence of basal NF-kB activity) that lead to hepatocellular carcinoma. Blocking Necroptosis (RIP3) or NF-kB-(IKKÃ) activation in those mice blocked hepatocarcinogenesis. Overall design: WT, combined conditional deletion of Traf2 and Caspase-8 in hepatocytes (DKO mice) (T2), inter-bred DKO mice with constitutive RIPK3 deficient mice RIP3-TKO(RIP3), interbred DKO mice with IKKÃ-floxed mice to facilitate hepatocyte specific triple knockout mice IKKÃ-TKO(IKK2)
