The transcription factor Mac1 is a key regulator of copper homeostasis and controls the transcriptional response to copper-limiting conditions in fungi. Expression analyses performed in the soil-borne plant pathogen Fusarium oxysporum revealed that almost all copper starvation-induced genes are downregulated in the absence of the regulator Mac1. The aim of this ChIP-seq analysis is to elucidate which of these genes are direct targets of Mac1. Overall design: Identification of Mac1-DNA interactions under copper limiting conditions and presence of copper in Fusarium oxysporum f. sp. lycopersici 4287 by ChIP-seq analysis. A mac1Stag mutant, expressing a version of Mac1 protein with the oligopeptide S-tag fused to the C-terminus, was constructed for the analysis. The S-tag was used as the epitope for the antibody a-S-tag (SAB2702227, Sigma-Aldrich®). Wt samples were used as negative control in the immunoprecipitacion assay. Three biological replicates of mac1Stag mutant were used.
