Our study provides considerable gene expression information of Trichophyton mentagrophytes at the transcriptional level, which will help accelerate the research on the antifungal medicine development. Additionally, we have demonstrated the feasibility of using the Illumina sequencing based DGE system for gene expression profiling, and have shed new light on functional studies of the genes involved in antifungal mechanisms of berberine hydrochloride and clotrimazole. Overall design: Berberine hydrochloride (BC) (HPLC > 98 %, Lot Number:20130306) was purchased from Yuan Ye Biological technology co., Ltd, Shanghai, China. The BC treated group (three samples) was subject to medicinal stress dose which means 1/2 MIC (minimum inhibitory concentration) dose (1 mg/mL berberine in DMSO), Clotrimazole (CLO) (99 % pure, Lot No. 23593-75-1) was purchased from BaDaTong Medical Company (TaiZhou, Zhejiang Province, China). And clotrimazole treated group (three samples) was subject to medicinal stress dose which means 1/2 MIC (minimum inhibitory concentration) dose (0.5 µg/mL Clotrimazole in DMSO),while the control group was fed with DMSO (Purity>99%) which was purchased from Sangon Biotech co., Ltd Shanghai, China only. Each group has three repeats. After 6 h incubation (28°C,150 rpm), the seeds were filtered with gauze and frozen after treatment for RNA isolation and further analysis.
