The study was conducted to understand the mechanism and therapeutic effect of human bone marrow derived mesenchymal stem cell in Crohn's disease using the SAMP1/YitFc (SAMP) mice, a spontaneous murine model of Crohn's like-ileitis. SAMP mice with established inflammation were treated with five million human bone marrow derived mesenchymal stem cells to study the therapeutic potential of MSCs. Mouse were euthanized at the endpoints, and mesentery was harvested and processed for the isolation of cells. Epithelial cells from inflammation-free young SAMP and control mice served as controls. Harvested cells were fixed using a fixation kit from Parse Bioscience, and single-cell RNA sequencing was performed using the SPLit-Seq approach from the Parse Bioscience platform to study the cellular phenotypes and transcriptomic signature of various cell types during diseased and MSCs treatment conditions.
