This dataset combines transcriptional and chromatin analysis of neutrophils from the air pouch mouse model, an in vivo model of acute inflammation, with transcriptional analysis of Hoxb8 neutrophils, an in vitro model of neutrophil development. Overall design: At 6 days after the creation of the air pouch, mice were challenged with 100 µg zymosan injected directly into the pouch. Four hours after zymosan injection the bone marrow, blood, membrane, and exudate were harvested for ATAC-sequencing and RNA-sequencing analysis. Additionally, we used an ex vivo system of murine ER-Hoxb8 neutrophils that phenotypically and morphologically recapitulate neutrophil maturation stages during five days of in vitro culture in the presence of G-CSF. The Hoxb8 cells of day 0, 1, 3, or 5 of development with or without 2 hours of incubation with zymosan were prepared for RNA-sequencing analysis.
