Chronic kidney disease (CKD) affects more than 10% of the global population with limited effective therapies. Understanding the underlying mechanisms of kidney fibrosis, the common final pathway of CKD to kidney failure, is essential to the development of new therapeutic strategies. Activation of the complement system and excessive production of the anaphylatoxin C5a observed in CKD suggests its possible role in disease pathogenesis and progression by promoting inflammation-driven kidney injury. This study aims to investigate the diverse roles of C5a receptor 1 (C5aR1) in two models of progressive CKD and provide evidence on C5aR1 antagonism as a therapeutic strategy.
慢性肾脏病(CKD)影响着全球超过10%的人口,且有效治疗方法有限。深入了解肾脏纤维化这一CKD向肾脏衰竭发展的共同终末途径的潜在机制,对于新疗法的研发至关重要。在CKD中观察到的补体系统的激活和过敏毒素C5a的过量产生,暗示了其在疾病发病机制和进展中可能发挥的作用,即通过促进炎症驱动的肾脏损伤。本研究旨在探究C5a受体1(C5aR1)在两种CKD进展模型中的多种作用,并为C5aR1拮抗剂作为治疗策略提供证据。
