AFM images of bare DNA origami rectangles on mica that were assembled at different staple excess (2x to 10x). Images .jpk were recorded in air using a JPK Nanowizard Ultra Speed (JPK Instruments) operated in intermittent contact mode with HQ:NSC/Al BS cantilevers (75 kHz, 2.8 N/m) from MikroMasch (NanoAndMore). Images .spm were recoreded in air using a Bruker Dimension ICON in ScanAsyst PeakForce Tapping mode with SCANASYST-AIR cantilevers (70 kHz , 0.4 N/m).
AFM images of biotinylated DNA origami rectangles after up to five (1b to 5b) consecutive folding reactions with staples recovered fromt he previous folding reactions. The DNA origami rectangles were adsorbed on mica and subsequently exposed to streptavidin (215 µM, 30 min) to visualize the presence of the biotinylated staples. Images were recorded in air using a Bruker Dimension ICON in ScanAsyst PeakForce Tapping mode with SCANASYST-AIR cantilevers (70 kHz , 0.4 N/m).
