A Fosmid library was prepared from NA12878 genomic DNA, using the pFosill-4 cloning vector. The library was grown in E. coli. Colonies were plated out and picked by hand. About 50 clones were selected, pooled, and propagated in liquid culture. This was repeated, yielding two pools of ~50 clones. The resulting Fosmid DNA was isolated, sheared to ~500 bp and equipped with Illumina sequencing adapters prior to end sequencing on MiSeq to 250 bases, with one lane for each pool. Aliquots of Fosmid DNA from the two pools were then combined and used as input for the creation of a jumping library (then sequenced on MiSeq), and a Pacific Biosciences library, which was sequenced, yielding reads of ~3000 bases. These data were assembled, yielding in all cases single contigs. Some loci in these contigs are undergoing laboratory validation and may be edited to reflect possible corrections.
