Effect of Notch3 perturbation on healthy and arthritic mouse synovium. Overall design: Mouse synovial tissues were dissected from knee joints. Pooled synovial tissue from 10 mice from each treatment group were enzymatically dissociated. Synovial cells were stained with LIVE/DEAD stain (ThermoFisher) and live synovial cells were isolated by flow cytometric sorting. 7,000 - 10,000 live synovial cells were then used for droplet-based single cell RNAseq (10X Genomics). Mice were either healthy or arthritic (serum transfer from K/BxN). Treatment conditions: isotype control vs anti-Notch3 mAB, wild type vs Notch3-/- knock out.
