We designed CRISPR-Cas9 whole genome screens to identify and rank critical genes for cholesterol metabolism in modified LDL treated BMDMs Overall design: Identify and rank critical genes for cholesterol metabolism in bone marrow-derived macrophages (BMDM) by CRISPR/Cas9 whole genome screen. The screens assayed cellular neutral lipid amounts following exposure of BMDM to low-density lipoprotein. Here, we have screens with change in neutral lipid content following 1) 24-h exposure to AcLDL or OxLDL (24h LDL) 2) 24-h exposure followed by 36-48 h post AcLDL or OxLDL removal culture (post-LDL 48h)
