Effect of EZH2 inhibition and CART19 on gene expression of B-NHL cell lines (OCI-ly18 and SU-DHL-4)

The goas of this experiment were: 1) to assess the effect of tazemetostat on B-NHL tumor cells lines; 2) to assess the effect of CART19 killing on B-NHL tumor cells lines; 3) to assess the effect of combining tazemetostat/CART19 on B-NHL tumor cells lines. OCI-ly18-GFP+ or SU-DHL-4-GFP+ were pre-treated for 3 days with tazemetostat (500 nM); killing assay was performed by adding CART19 vs UTD cells (+/- tazemetostat). After 48 hours, tumor cells were sorted from 4 experimental conditions (UTD-DMSO, UTD-TAZ, CART19-DMSO, CART19-TAZ) and RNA was extracted for bulk RNA sequencing.
