Genomic DNA engineering is considered one of the promising techniques for designing new functional bacterial genomes for future industrial applications. However, there is limited information available regarding structurally labile genomic regions or sequences during long-term cultivation. To assess such global structural changes or mutational changes of the bacterial genome during long-term cultivation, two lineages of long-term cultured Escherichia coli ATCC 25922 genomes were re-sequenced independently. The global structural changes were monitored by a long-read sequencing with the Oxford Nanopore sequencing and the single nucleotide level of mutations were detected by a short-read sequencing with the DNBSEQ. The completeness of the re-sequenced two genomic DNAs was verified with CheckM.
