Regulatory T (Treg) cells, which specifically express the transcription factor Foxp3, are actively engaged in life-long suppression of potentially hazardous self-reactive T cells to prevent autoimmune disease. The majority of them develop in the thymus as a functionally distinct and mature T-cell subpopulation, with their Foxp3 expression stabilized mainly by Treg-specific DNA demethylation of the Foxp3 enhancer region CNS2. In this study, we characterized the molecular mechanisms underlying thymic Treg cell development.
