Sequencing of root transcriptomes from wilt-susceptible ICP 2376 and wilt-resistant ICP 8863 pigeonpea cultivars obtained during early stage of Fusarium udum infection.

Pigeonpea [Cajanus cajan (L.) Millspaugh] is the seventh most important legume crop and global productivity of pigeonpea is nearly 4.43 million tonnes. Fusarium udum Butler is the most important pathogen causing the devastating vascular wilt disease of this grain legume. Yield of pigeonpea is severely curtailed by this fungal pathogen and total annual loss has been estimated at 470000 t in India. Present study was conducted by inoculating wilt-susceptible ICP 2376 and wilt-resistant ICP 8863 pigeonpea cultivars with F. udum and was compared with their non-inoculated counterparts. Total mRNA was collected from all four candidate plant root samples at 36 hours post inoculation (early time point of infection) and transcriptomes were sequenced using next generation sequencing technique. Adequate amount of high quality reads were generated with a maximum read length of 150 bp. Total number of raw reads are 28.39 m, 28.31 m, 35.74 m and 26.25 m for non-inoculated resistant (NIR), non- inoculated susceptible (NIS), inoculated resistant (IR) and inoculated susceptible (IS), respectively. Processed read counts revealed that 27.2 m, 26.95 m, 34.35 m and 25.36 m reads were obtained from NIR, NIS, IR and IS, respectively. De novo assembly processed data was performed using Trinityrnaseq_r20140717 and COGs has been used for further analysis. Processed transcripts were annotated using NCBI blastn 2.303 with the DNA and protein sequences of Cajanus cajan, Glycine max, Phaselous vulgaris, Vigna radiata, Lotus Japonicas, Medicago truncatula and Arabidopsis thaliana. Assembly of processed reads into transcripts generated 322688 in total, with maximum transcript length of 24582 bp and minimum length of 300 bp. Total transcript length was found to be 29,58,30,572 bp with 0% non ATGC characters.
