Single cell analysis of newly diagnosed AML patients

Acute Myeloid Leukemia (AML) is a heterogenous disease characterized by immature blasts at different maturation stage. We used single cell sequencing technique to analyze newly diagnosed AML patients. Overall design: Bone marrow biopsies were routinely collected before treatment initiation. Samples were freshly frozen with freezing medium containing 20% fetal calf serum (FCS) and 10% dimethyl sulfoxide (DMSO) in Dulbecco's modified Eagle medium (DMEM) and stored in liquid nitrogen. All frozen BM samples were retrieved immediately before sample processing. The 5 prime gene expression libraries were prepared using a 10x Genomics Chromium Controller instrument and Chromium Single-Cell 5 V5.1 reagent kits (10x Genomics).
