These data are intended for the storage of sequencing data in FASTQ format within the project entitled Application of nanopore sequencing of activated sludge bacteriome for wastewater treatment process control.The aim of this project is the detection of technologically undesirable bacterial species occurring in activated sludge using nanopore sequencing. The objective was to design and optimize a nanopore sequencing method targeting the 16S rRNA gene to determine bacterial diversity and subsequently detect and quantify significant bacterial species that influence the proper course of the wastewater treatment process. The dataset consists of a total of six files containing raw data in FASTQ format, serving as input data for the project output TQ03000804 in the form of a Ztech-verified technology. The measurements were carried out in parallel at two workplaces, namely VŠCHT Prague and PVK, a.s., in three temporally separated sequencing runs (i.e., three files for the PVK, a.s. laboratory and three files for VŠCHT Prague). The data were subjected to so-called basecalling, during which the data were converted from POD5 format to FASTQ format. This process involves assigning specific nucleotide bases to individual changes in electrical current using the MinKNOW software (Oxford Nanopore Technologies). In the subsequent step, the data must undergo demultiplexing based on barcode sequences specific to individual samples, along with the removal of sequencing adapters, barcodes, and primers (trimming).
