Nascent transcription measured by GRO-seq in Zea mays B73 seedlings

Global run-on sequencing (GRO-seq) libraries were each made from 10 pooled day-6 aerial Zea mays seedlings in the B73 inbred. Nuclei were isolated and nascent transcripts were labelled by incorporation of 5-ethynyl-UTP which was biotinylatated by Click-iT reaction. Biotinylated RNAs were captured on streptavidin beads and libraries were prepared using the BIOO Scientific NEXTFLEX Small RNA-Seq Kit v3 with no size selection.
