Deep mutational scanning of complex I assembly factor NDUFAF6

The goal of this study is to assess the function of single amino acid missense mutations in the complex I assembly factor NDUFAF6. NDUFAF6 knockout cells were transduced with a library of NDUFAF6 variants. Cells expressing non-functional variants of NDUFAF6 were selected against by culturing cells in media containing galactose as the primary carbon source. Cells pellets were collected before and after the galactose selection for gDNA isolation and NGS sample prep.
