The carnitine palmitoyltransferase (CPT) enzyme family regulates long-chain fatty acid oxidation and represents an important therapeutic target in metabolic diseases such as obesity, diabetes, and cancer. CPT1A and CPT1B are located in the outer mitochondrial membrane, where they convert fatty acids into acyl-carnitines that are transported into mitochondria to fuel β-oxidation. Although CPT isoforms share a common catalytic fold, they differ in localization, regulation, and function. Thus, structural information on CPT1 isoforms is largely missing. Therefore, structurally characterizing CPT1A in complex with physiological ligands and malonyl-CoA inhibitor using cryoEM is essential to understand CPT regulation and support therapeutic development.
