Extrachromosomal circular DNA (eccDNA) is known for its contribution to genome dynamics. eccDNA is proposed to be generated from DNA ligation. Eukaryotic genomes are occupied by abundant retrotransposons, however, due to the lack of efficient tools, whether retrotransposons can form eccDNA upon activation is largely unknown. Here, by applying Nanopore sequencing, we examine retrotransposon mobilization and eccDNA formation.
