Genome sequencing of Psychotria manillensis Bartl. ex DC. was performed by using Roche 454 reads. Total genomic DNA of P. manillensis was extracted from dried leaf tissue collected from Okinawa Island (Sueyoshi-Park, Naha City; 26?13'38.0"N 127?42'54.5"E), using the DNasy Plant Mini Kit. The extracted DNA was pyrosequenced using a 454 GS Junior System. Multiplex Identifier (MID) tags were used for multiplexing of this sample and other samples. These samples were combined. The row data were demultiplexed and MID tags removed from the reads using the Newbler software package.
