Brain transcriptome analysis in the cortex and hippocampus of Cyfip2 Arg87Cys knock-in mice at four different postnatal stages

To investigate age-dependent molecular changes in the cortex and hippocampus of Cyfip2 Arg87Cys knock-in mice, we conducted transcriptome analysis at postnatal weeks 1, 7, 14, 28. We dissected the frontal cortex and hippocampal regions from both hemispheres of each mouse. WT and Cyfip2 Arg87Cys knock-in mice were littermates, with four mice (replicates) prepared for each age-genotype group. After dissection, tissue samples were stored in RNAlater at 4C overnight and then at -80C before being processed further for RNA extraction.
为探究Cyfip2 Arg87Cys敲入小鼠(Cyfip2 Arg87Cys knock-in mice)皮层与海马体的年龄依赖性分子变化,我们于产后第1、7、14、28周开展转录组分析(transcriptome analysis)。我们从每只小鼠的双侧大脑分离额叶皮层与海马组织区域。野生型(Wild Type, WT)与Cyfip2 Arg87Cys敲入小鼠均为同窝仔鼠,每个年龄-基因型组设置4只小鼠作为生物学重复。组织分离完成后,样本先置于RNAlater溶液中4℃静置过夜,随后转移至-80℃保存,直至后续进行RNA提取实验。
