The CRISPR screen in mice was conducted to identify gene(s) involved in the apoptotic death of immune cells. Mice were reconstituted with a CRISPR library (Koike-Yusa, et al. Nat Biotechnol 32, 267-273 (2014)) and subjected to polymicrobial sepsis. The surviving cells in the bone marrow were isolated and subjected to next gen sequencing to identify Treml4 as the gene that regulates innate immune cell death during sepsis.
