Transcriptome sequencing of Symbiodinium minutum

In order to obtain a diverse pool of mRNA transcripts, S. minutum cultures which are usually maintained at 25 ?C were subjected to either short-term (3 hrs) or long-term (24 and 72 hrs) high temperature stress (31?C) under the same light condition. We prepared seven libraries including controls (0, 3, 24, and 72 hrs) and heat-stressed samples (3, 24, and 72 hrs). Illumina 75-bp paired-end RNA-seq sequencing was performed, all high-quality sequences obtained from the seven libraries were assembled with Velvet/Oases assembler with hash length 37.
