The present study was designed to investigate the effect of Endothelin-1 (ET-1) on human GV-stage oocyte maturation and explore the mechanism of ET-1 induced resumption of meiosis. Adding ET-1 to the culture medium may enhance the occurrence rate of germinal vesicle breakdown (GVBD) in cumulus cell-enclosed human oocytes. We used whole-genome transcriptome microarray to analysis the differential expression gene in cumulus cells Overall design: Two samples were analyzed. Cumulus cells from GV-stages COCs cultured in G-IVF medium for 18 hours, with or without 100ng/ml ET-1. As a better control, we harvest cells at the same time. The sample 18Hmean GV-stages cumulus cells treated with ET-1 medium for 18 hours. The sample 0H mean GV-stages cumulus cells treated without ET-1 medium for 18 hours.
