叶绿素是海洋浮游植物中的光合色素,其含量是体现海洋中浮游植物以及初级生产力分布的重要指标,同海洋碳汇、海洋中营养物质以及渔业资源的分布有着紧密的联系。 本数据为2016年中国第7次北极考察叶绿素a观测数据。中国第七次北极考察期间共完成82个站位叶绿素采样观测。共获取叶绿素样品594份,其中粒度分级叶绿素样品498 份(分大于20 μm,2?20 μm,小于2 μm 3个粒级进行分析测定),真光层总叶绿素样品96份,现场分析取得叶绿素数据1590个。 叶绿素a的测定采用萃取荧光法。使用干净取样瓶在规定站位和层次采取水样,水样收集前,经200 μm 孔宽的筛绢预过滤,以除去大多数的浮游动物。采样层次按标准层。过滤250 cm3水样,色素用90% 丙酮萃取24 h,用唐纳荧光计进行测定。分级叶绿素a 水样经孔宽20 μm的筛绢、孔宽2.0 μm 的Nuclepore 滤膜和Whatman GF/F 玻璃纤维滤膜过滤,以分别获取网采(Net 级份,>20 μm)、微型(Nano级份,2?20 μm)和微微型(Pico级份, 0.2 ?2 μm)的光合浮游生物,具体测定方法与叶绿素a相同。
Chlorophyll is a photosynthetic pigment in marine phytoplankton. Its concentration serves as a critical indicator for the distribution of marine phytoplankton and marine primary productivity, and is closely correlated with the distributions of marine carbon sinks, marine nutrients and fishery resources. This dataset contains chlorophyll a observation data collected during the 7th Chinese National Arctic Research Expedition in 2016. A total of 82 stations were sampled for chlorophyll during the expedition, and 594 chlorophyll samples were obtained in total. Among them, 498 were size-fractionated chlorophyll samples, which were analyzed and measured in three size classes: >20 μm, 2–20 μm, and <2 μm; additionally, 96 total chlorophyll samples from the euphotic zone were collected. A total of 1590 chlorophyll data points were obtained through in-situ analysis. Chlorophyll a was determined using the extraction-fluorometry method. Water samples were collected using clean sampling bottles at designated stations and depths. Prior to sample collection, water was pre-filtered through a 200 μm mesh sieve to remove most zooplankton. Sampling depths followed standard depth layers. A 250 cm³ water sample was filtered, and pigments were extracted with 90% acetone for 24 hours, followed by measurement using a Turner fluorometer. For size-fractionated chlorophyll a samples, water was filtered through a 20 μm mesh sieve, a 2.0 μm Nuclepore membrane, and a Whatman GF/F glass fiber filter, respectively, to collect net (>20 μm), nano (2–20 μm), and pico (0.2–2 μm) photosynthetic plankton. The specific determination methods were identical to those used for total chlorophyll a.
