A 5.6 cM genetic map of S. mansoni had been generated by the Anderson and LoVerde labs (Criscione et al., 2009). To identify drug resistant genes using the map, a male parent from the drug resistant strain was crossed with a female parent from a drug sensitive strain. The oxamniquine drug resistant strain was established in the 1970âs and the drug sensitive strain is the LE strain from Brazil. The LE strain was one of the parents used to generate the genetic map. Eggs from the cross (F1 progeny) were used to infect snails with a single miracidia (single genotype) and the resulting cercariae sexed. A F1 male and a F1 female were crossed to produce F2 progeny. Three thousand snails were exposed to a single miracidium. Of the 3000 exposed over 600 were positive for infection. The F2 adult progeny were phenotyped for drug sensitivity in an in vitro assay. The parents, F1s and F2s are being genotyped. Sequencing the single genotypes the information contributes to the assembly (the current draft was generated from a population of parasites) and will produce a SNP map.
