Intestinal Cyp24a1 regulates vitamin D locally independent of systemic regulation by renal Cyp24a1 in mice

Vitamin D is a central regulator of mineral homeostasis, but its cell-type-specific effects are poorly understood. Here, we tested the effects of acute calcitriol injection on gene expressions in mouse kidneys and intestine (jejunum) using single-cell RNA sequencing. To evaluate the difference in gene expressions between control mice and vitamin D-treated mice in each cell type, we performed single-cell transcriptomic analyses with 10x Chromium 3' v3.1 using dissociated total kidney cells and intestinal cells (duodenum, 1:1 for villus and crypt-derived cells).
