RNA seq with AML cells (NB4) upon FTO knockdown. Homo sapiens

To identify the potential targets genes of FTO in acute myeloid leukemia (AML), we peroformed RNA-seq of NB4 cells with or without FTO knockdown based on Illumina Hiseq system. Overall design: NB4 cells were infected with pLKO.1-shFTO and pLKO.1-shNS lentivirus and the positive cells were selected with 1ug/ml puromycin 72 hours post lentivirus infection. After two generation of selections, the NB4 cells were harvested and RNA samples were extracted for sequencing.
