Knee osteoarthritis (KOA) is a degenerative joint disease, and pain is the main reason patients with osteoarthritis seek medical care. Bushen Zhuangjin Decoction (BZD), as a traditional Chinese medicine for the treatment of osteoarthritis, has good anti-inflammatory and analgesic effects. However, whether BZD can affect the brain function activity of KOA patients to treat pain has not been reported. In this study, we selected the traditional Chinese medicine database TCMSP, and major disease databases included GeneCards database, OMIM database, PharmGkb database, TTD database, DrugBank database, and GEO gene chip database. GeneCards database, OMIM database, PharmGkb database, TTD database,and DrugBank database all used osteoarthritis as the search term. All of the categories were selected for humans to obtain differential genes for human osteoarthritis. In order to make the results more comprehensive and reliable, the collection of these five databases is selected here as A. GEO database was searched with osteoarthritis and subchondral bone as keywords, and human was selected as the category. Here is the differential gene of human osteoarthritis subchondral bone, and here is B. The intersection of A and B is the differential gene of subchondral bone in human osteoarthritis. TCMSP was used to screen the effective compounds and targets of BZD. Combination of disease target and drug target to predict the possible target of BZD in the treatment of KOA pain. Then in vivo tests showed that BZD could reduce inflammation and relieve pain by altering nerve signaling. Finally, fMRI was used to verify that BZD treatment of KOA is related to central nervous regulation.
