The study aims to show that a metabolic shift is a precondition for the initiation of an invasive/metastatic program in pancreatic cancer. Paracrine cues and metabolic stress (mimicked in vitro by treatment with macrophage-conditioned media and low dose etomoxir) regularly found in the metastasis-promoting tumor stroma consistently induced transcriptional changes resulting in the acquisition of migratory and invasive abilities. Samples from primary cultures corresponding to 3 different patient-derived xenografts either untreated (CONT, used as reference for each Pdx) or treated for 48h with macrophage conditioned media (MCM) or low-dose etomoxir (ETO), in duplicate.
