Mature eggs enclose maternal messengers RNA necessary to initiate a new life, and therelated RNA transcripts provide a means of protein synthesis without transcriptionalprocess, during early embryogenesis, prior to the zygotic gene expression. TheNeotropical catfish Pseudopimelodus mangurus is an endangered species in whichconservation efforts are necessary. Reproductive biotechniques for generation of agermline chimera is a tool that can be applied to the conservation and subsequentreconstitution. Primordial germ cells (PGC) are able to transmit the genetic informationfor progenies and therefore, PGC may be used for genebanking and transplantation insterile recipients. The identification of PGC during the early stages is an essential stepin such a transplantation and traceability of PGCs is achieved by microinjection of RNAof the coding region of reporter gene in fusion with 3’-untranslated region of thespecific PGC cDNA. The mechanism of PGC specification is common in fish and otherorganisms maternally inherited cytoplasmic determinants is believed to give rise to thegerm lineage. The egg transcriptome of the Pseudopimelodus mangurus will permitthen to identify the maternal RNAs involved in the different metabolic pathways,allowing the investigation of the molecular mechanisms related with differentiation andmigration of PGC.
